New knowledge about the responses of species to fire is needed to plan for biodiversity conservation in the face of changing fire regimes. However, the knowledge that is acquired may be influenced by the sampling method and the taxonomic resolution of a study. To investigate these potential sampling biases, we examined invertebrate responses to time since fire in mallee woodlands of southern Australia. Using a large‐scale replicated study system, we sampled over 60 000 invertebrates with large pitfall traps, wet pitfall traps and sweep nets, and undertook analyses at morphospecies and order level. Large pitfalls and sweep nets detected several strong fire effects, whereas wet pitfall traps detected few effects. Invertebrate abundance in sweep nets was highest shortly after fire because of grasshopper outbreaks. Several additional morphospecies showed strong preferences for different stages in the post‐fire succession. In contrast with morphospecies effects, analyses at order level either failed to detect fire effects or were driven by the most abundant species. For fire research to produce credible results with the potential to guide management, it must use a range of sampling techniques and undertake analyses at (morpho)species level. Homogeneous fire management, such as fire suppression in fragmented landscapes or widespread frequent burning for asset protection, is likely to cause declines in fire‐affected invertebrates.  相似文献

Free radical activity towards superoxide anion radical (), hydroxyl radical (HO?) and 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH?) of a series of novel thiazolidine‐2,4‐dione derivatives (TSs) was examined using chemiluminescence, electron paramagnetic resonance (EPR) and EPR spin trapping techniques. 5,5‐Dimethyl‐1‐pyrroline‐N‐oxide (DMPO) was applied as the spin trap. Superoxide radical was produced in the potassium superoxide/18‐crown‐6 ether dissolved in dimethyl sulfoxide. Hydroxyl radical was generated in the Fenton reaction (Fe(II) + H2O2. It was found that TSs showed a slight scavenging effect (15–38% reduction at 2.5 mmol/L concentration) of the DPPH radical and a high scavenging effect of (41–88%). The tested compounds showed inhibition of HO? ‐dependent DMPO‐OH spin adduct formation (the amplitude of EPR signal decrease ranged from 20 to 76% at 2.5 mmol/L concentration. Significantly increased power conversion efficiency (PCE) of polymer solar cells (PSCs) is achieved by applying a plasmonic enhanced light trapping strategy to a low bandgap conjugated polymer, poly(indacenodithiophene‐ co‐phananthrene‐quinoxaline) (PIDT‐PhanQ) and [6,6]‐phenyl‐C71‐butyric acid methyl ester (PC71BM) based bulk‐heterojunction (BHJ) system. By doping both the rear and front charge‐selecting interfacial layers of the device with different sizes of Au NPs, the PCE of the devices is improved from 6.65% to 7.50% (13% enhancement). A detailed study of processing, characterization, microscopy, and device fabrication is conducted to understand the underlying mechanism for the enhanced device performance. The success of this work provides a simple and generally applicable approach to enhance light harnessing of low bandgap polymers in PSCs.  相似文献

The capsid (CA) protein plays crucial roles in HIV infection and replication, essential to viral maturation. The absence of high-resolution structural data on unassembled CA hinders the development of antivirals effective in inhibiting assembly. Unlike enzymes that have targetable, functional substrate-binding sites, the CA does not have a known site that affects catalytic or other innate activity, which can be more readily targeted in drug development efforts. We report the crystal structure of the HIV-1 CA, revealing the domain organization in the context of the wild-type full-length (FL) unassembled CA. The FL CA adopts an antiparallel dimer configuration, exhibiting a domain organization sterically incompatible with capsid assembly. A small compound, generated in situ during crystallization, is bound tightly at a hinge site (“H site”), indicating that binding at this interdomain region stabilizes the ADP conformation. Electron microscopy studies on nascent crystals reveal both dimeric and hexameric lattices coexisting within a single condition, in agreement with the interconvertibility of oligomeric forms and supporting the feasibility of promoting assembly-incompetent dimeric states. Solution characterization in the presence of the H-site ligand shows predominantly unassembled dimeric CA, even under conditions that promote assembly. Our structure elucidation of the HIV-1 FL CA and characterization of a potential allosteric binding site provides three-dimensional views of an assembly-defective conformation, a state targeted in, and thus directly relevant to, inhibitor development. Based on our findings, we propose an unprecedented means of preventing CA assembly, by “conformationally trapping” CA in assembly-incompetent conformational states induced by H-site binding.  相似文献

Mating speed and copulation duration respond rapidly to laboratory selection in Drosophila melanogaster Meigen (Diptera: Drosophilidae), but there is a lack of data on the evolutionary response to natural selection in the wild. Further, it is not clear whether body melanization and mating behavior are correlated traits. Accordingly, we tested whether variation in body color impacts on mating latency, copulation duration, and fecundity in latitudinal populations of D. melanogaster. We observed geographical variation (cline) for mating propensity, i.e., mating speed as well as copulation duration increased along latitude. Phenotypic plastic responses for body melanization at 17 and 25 °C also showed significant correlations with mating latency and copulation duration. Within‐population analysis based on assorted dark and light flies of five geographical populations showed significant positive correlations of copulation duration and fecundity with body melanization. To assess the role of males and/or females on mating speed and copulation duration, we used atypical body color strains (i.e., dark and light males of D. melanogaster) for no‐choice mating tests. Our data showed a major influence of males for copulation duration and of females for mating speed. Furthermore, a difference in impact of body melanization on mating speed and copulation duration was demonstrated between species, i.e., low melanization in Drosophila ananassae Doleschall is correlated with lower mating speed and shorter copulation duration than in D. melanogaster. Geographical changes in mating propensity were significantly correlated with body melanization at three levels, i.e., within and between populations and between species. Thus, we have shown that a relationship exists between body melanization and mating success. Further, we found seasonal changes in temperature and humidity to confer selection pressures on mating‐related traits.  相似文献

Various techniques exerting mechanical stress on cells have been developed to investigate cellular responses to externally controlled stimuli. Fundamental mechanotransduction processes about how applied physical forces are converted into biochemical signals have often been examined by transmitting such forces through cells and probing its pathway at cellular levels. In fact, many cellular biomechanics studies have been performed by trapping (or immobilizing) individual cells, either attached to solid substrates or suspended in liquid media. In that context, we demonstrated two‐dimensional acoustic trapping, where a lipid droplet of 125 µm in diameter was directed transversely toward the focus (or the trap center) similar to that of optical tweezers. Under the influence of restoring forces created by a 30 MHz focused ultrasound beam, the trapped droplet behaved as if tethered to the focus by a linear spring. In order to apply this method to cellular manipulation in the Mie regime (cell diameter > wavelength), the availability of sound beams with its beamwidth approaching cell size is crucial. This can only be achieved at a frequency higher than 100 MHz. We define ultrasound beams in the frequency range from 100 MHz to a few GHz as ultrasound microbeams because the lateral beamwidth at the focus would be in the micron range. Hence a zinc oxide (ZnO) transducer that was designed and fabricated to transmit a 200 MHz focused sound beam was employed to immobilize a 10 µm human leukemia cell (K‐562) within the trap. The cell was laterally displaced with respect to the trap center by mechanically translating the transducer over the focal plane. Both lateral displacement and position trajectory of the trapped cell were probed in a two‐dimensional space, indicating that the retracting motion of these cells was similar to that of the lipid droplets at 30 MHz. The potential of this tool for studying cellular adhesion between white blood cells and endothelial cells was discussed, suggesting its capability as a single cell manipulator. Biotechnol. We set a twofold investigation: we assess left ventricular (LV) rotation and twist in the human heart through 3D-echocardiographic speckle tracking, and use representative experimental data as benchmark with respect to numerical results obtained by solving our mechanical model of the LV. We aim at new insight into the relationships between myocardial contraction patterns and the overall behavior at the scale of the whole organ. It is concluded that torsional rotation is sensitive to transmural gradients of contractility which is assumed linearly related to action potential duration (APD). Pressure-volume loops and other basic strain measures are not affected by these gradients. Therefore, realistic torsional behavior of human LV may indeed correspond to the electrophysiological and functional differences between endocardial and epicardial cells recently observed in non-failing hearts. Future investigations need now to integrate the mechanical model proposed here with minimal models of human ventricular APD to drive excitation-contraction coupling transmurally.  相似文献

西双版纳山地橡胶林凋落物的生态水文效应   总被引:1,自引:0,他引:1  
森林凋落物具有阻滞径流、拦截泥沙和均衡土壤水热状况等功效.以西双版纳地区20年生橡胶林为对象,采用径流微小区法和剔除-对照凋落物等实验,测定了其凋落物的生态水文效应,结果表明:橡胶林坡地剔除凋落物径流微小区(2009年10月-2010年9月)径流量为625.7±18.5 mm,显著高于对照的185.1±72.7 mm;剔除凋落物径流泥沙浓度与最大1h降雨强度呈显著正相关.剔除凋落物使橡胶林坡地(6、7月除外)和台地(8月除外)表层土壤(0~10 cm)湿度均显著降低;剔除凋落物表土平均温度在雾凉季(11月-翌年2月)显著低于对照,而在干热季(3-4月)显著高于对照,且剔除凋落物表土温度整年都比对照具有更大日变化幅度.可见橡胶林凋落物具有较好的生态水文功效,然而,受频繁人为割胶活动等的影响,台地凋落物的分解速率在雨季中、后期加快,不利于其生态水文效应的发挥,故在营林活动和生态恢复中应加强对凋落物的管理与保护.  相似文献

The acquisition of hypsodont molars is often regarded as a key innovation in the history of ruminant ungulates. Hypsodont ruminants diversified rapidly during the later Neogene, circa 15-2 Myr ago, and came to dominate the ruminant fossil record in terms of species diversity. Here we show that hypsodont clades had higher speciation and diversification rates than other clades. Hypsodont species had, on average, shorter stratigraphic durations, smaller range size and lower occupancy than non-hypsodont species. Within hypsodont clades, some species were very common and acquired large geographical ranges, whereas others were quite rare and geographically limited. We argue that hypsodont clades diversified in an adaptive radiation-like fashion, with species often splitting cladogenetically while still in the expansive phase of their occupancy history.  相似文献
